Fibrinogen (clotting factor I) is a heat labile beta glycoprotein present in plasma. It is the precursor of fibrin, which is the key protein constituting the network of the blood clot. Thrombin converts fibrinogen to fibrin by limited proteolysis. Fibrin monomers polymerize to fibrin which is stabilized by cross-linking Fibrinogen is isolated from fresh plasma after removing prothrombin. Freund's complete adjuvant is used in the first step of the immunization procedure.
Adsorption
Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies cross-reacting with other plasma proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.
Conjugation Procedure
A proprietary technique for the binding to FITC is used, followed by several purification steps to remove free reactants and protein aggregates. After each step activity and specificity are tested in a variety of techniques. The conjugate is lyophilized to assur stability and long shelf life.
Reactivity Note
The antiSerum does not cross-react with any other component of Mouse plasma. Inter-species crossreactivity is a normal feature of antibodies to plasma proteins since they frequently share antigenic determinants. Cross-reactivity of this antiSerum has not been tested in detail.
Preservative
No preservative added, as it may interfere with the antibody activity.
Physicochemical Characteristics
IgG protein concentration 10mg/ml. Fluorescein/IgG protein molar ration (F/P) approximately 1.9. No foreign proteins added.
Fluorescent Marker
Fluorescein isothiocyanate isomer 1 (FITC). Excitation: 495 nm, emission: 515 nm.
Intended Uses
As reagent for the direct detection of fibrinogen in mouse cells, tissues and body fluids in immunofluorescence; as detection reagent in non-isotopic methodology and solid phase immunochemistry (eg. ELISA). This immunoconjugate is not pre-diluted. The optimum working dilution of each conjugate should be established by titration before being used. Excess labelled antibody must be avoided because it may cause high unspecific background staining and interfere with the specific signal. Working dilutions are usually between 1:20 and 1:80.
Preparation and Storage
The lyophilized conjugate is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C. It is reconstituted by adding 1 ml sterile distilled water, spun down to remove insoluble particles, divided into small aliquots, frozen and stored at or below -20°C. Prior to use, an aliquot is thawed slowly in the dark at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezingshould be avoided.

Uniprot Description

Function: Fibrinogen has a double function: yielding monomers that polymerize into fibrin and acting as a cofactor in platelet aggregation.

Subunit structure: Heterohexamer; disulfide linked. Contains 2 sets of 3 non-identical chains (alpha, beta and gamma). The 2 heterotrimers are in head to head conformation with the N-termini in a small central domain

Subcellular location: Secreted.

Domain: A long coiled coil structure formed by 3 polypeptide chains connects the central nodule to the C-terminal domains (distal nodules). The long C-terminal ends of the alpha chains fold back, contributing a fourth strand to the coiled coil structure.

Post-translational modification: Conversion of fibrinogen to fibrin is triggered by thrombin, which cleaves fibrinopeptides A and B from alpha and beta chains, and thus exposes the N-terminal polymerization sites responsible for the formation of the soft clot. The soft clot is converted into the hard clot by factor XIIIA which catalyzes the epsilon-(gamma-glutamyl)lysine cross-linking between gamma chains (stronger) and between alpha chains (weaker) of different monomers.
