Western Blot (WB)
(Western blot analysis of EGLN2 Antibody (Center) in MDA-MB231, Hela cell line lysates (35ug/lane). EGLN2 (arrow) was detected using the purified Pab.)
Immunohistochemistry (IHC)
(Formalin-fixed and paraffin-embedded human testis tissue reacted with EGLN2 Antibody (Center), which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated.)
Flow Cytometry (FC/FACS)
(EGLN2 Antibody (Center) flow cytometric analysis of MDA-MB231 cells (right histogram) compared to a negative control cell (left histogram).FITC-conjugated goat-anti-rabbit secondary antibodies were used for the analysis.)
NCBI and Uniprot Product Information
NCBI Description
The hypoxia inducible factor (HIF) is a transcriptional complex that is involved in oxygen homeostasis. At normal oxygen levels, the alpha subunit of HIF is targeted for degration by prolyl hydroxylation. This gene encodes an enzyme responsible for this post-translational modification. Alternative splicing results in multiple transcript variants. Read-through transcription also exists between this gene and the upstream RAB4B (RAB4B, member RAS oncogene family) gene. Uniprot Description
EGLN2: Cellular oxygen sensor that catalyzes, under normoxic conditions, the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins. Hydroxylates a specific proline found in each of the oxygen-dependent degradation (ODD) domains (N-terminal, NODD, and C-terminal, CODD) of HIF1A. Also hydroxylates HIF2A. Has a preference for the CODD site for both HIF1A and HIF2A. Hydroxylated HIFs are then targeted for proteasomal degradation via the von Hippel-Lindau ubiquitination complex. Under hypoxic conditions, the hydroxylation reaction is attenuated allowing HIFs to escape degradation resulting in their translocation to the nucleus, heterodimerization with HIF1B, and increased expression of hypoxy-inducible genes. EGLN2 is involved in regulating hypoxia tolerance and apoptosis in cardiac and skeletal muscle. Also regulates susceptibility to normoxic oxidative neuronal death. 2 isoforms of the human protein are produced by alternative initiation.

Product Notes
The EGLN2 egln2 (Catalog #AAA6294730) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The EGLN2, ID (EGLN2, EIT6, Egl nine homolog 2, Estrogen-induced tag 6, HPH-3, Hypoxia-inducible factor prolyl hydroxylase 1, Prolyl hydroxylase domain-containing protein 1) (PE) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's EGLN2 can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), FLISA, Immunohistochemistry (IHC), Western Blot (WB). FC/FACS: 1:10-50 FLISA: 1:1,000 IHC: 1:50-100 WB: 1:100-500 Applications are based on unconjugated antibody. Researchers should empirically determine the suitability of the EGLN2 egln2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "EGLN2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
