This product consists of purified IgG fraction of the above antiserum covalently coupled with the N-Hydroxysuccinimide ester of biotin under mild conditions to give a high degree of substitution. The product is formulated in a phosphate saline buffer (0.01M, pH 7.2) containing 0.1% sodium azide preservative. No stabilizing proteins have been added. Possible applications for this product include avidin and streptavidinamplification systems for immunohistochemistry, ELISA, fluorescence microscopy and immunoblotting. In addition, this product may be used in place of neat antiserum in almost any appropriate antibody-based technique. Protein concentration is 4-5 mg/ml. Recommended short term (<6 months) storage is liquid at 2-8 degree C. For longer term storage, aliquot and freeze. Unit size is 1.0 ml.

Function: Diphtheria toxin, produced by a phage infecting Corynebacterium diphtheriae, is a proenzyme that, after activation, catalyzes the covalent attachment of the ADP ribose moiety of NAD to eukaryotic elongation factor 2 (eEF-2). Fragment A is the catalytic portion responsible for enzymatic ADP-ribosylation of elongation factor 2, while fragment B is responsible for binding of toxin to cell receptors and entry of fragment A. Catalytic activity: NAD+ + peptide diphthamide = nicotinamide + peptide N-(ADP-D-ribosyl)diphthamide.
Enzyme regulation: Partially inhibited by 1,8-naphthalimide (NAP). Ref.6
