Immunofluorescene (IF)
(MBS9600458 staining Hela by IF/ICC. The sample were fixed with PFA and permeabilized in 0.1% Triton X-100, then blocked in 10% serum for 45 minutes at 25 degree C. The primary antibody was diluted at 1/200 and incubated with the sample for 1 hour at 37 degree C. An Alexa Fluor 594 conjugated goat anti-rabbit IgG (H+L) Ab, diluted at 1/600, was used as the secondary antibody.)
Immunohistochemistry (IHC)
(MBS9600458 at 1/100 staining Rat testis tissue by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the primary antibody at 4°C overnight. An HRP conjugated anti-Rabbit antibody was used as the secondary antibody.)
Western Blot (WB)
(Western blot analysis of extracts from Human Fetal heart, using DGKH Antibody. The lane on the left was treated with blocking peptide.)
Function: Phosphorylates diacylglycerol (DAG) to generate phosphatidic acid (PA). Plays a key role in promoting cell growth. Activates the Ras/B-Raf/C-Raf/MEK/ERK signaling pathway induced by EGF. Regulates the recruitment of RAF1 and BRAF from cytoplasm to membranes and their heterodimerization.
Subunit Structure: Isoform 1 forms homooligomers through the SAM domain. Isoform 1 is also able to form heterooligomers with SAM domain-containing isoforms of DGKD. Oligomerization of isoform 1 inhibits its catalytic activity. Interacts with RAF1 and BRAF.
Post-translational Modifications: Phosphorylated; does not inhibit catalytic activity.
Similarity: Belongs to the eukaryotic diacylglycerol kinase family.
