Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for LRRC17 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any LRRC17 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LRRC17 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LRRC17 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Capture antibody was pre-coated onto 96-well plates. And the biotin conjugated antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the target amount of sample captured in plate. Principle of the Assay: TSKU ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-TSKU antibody and an TSKU-HRP conjugate. The assay sample and buffer are incubated together with TSKU-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TSKU concentration since TSKU from samples and TSKU-HRP conjugate compete for the anti-TSKU antibody binding site. Since the number of sites is limited, as more sites are occupied by TSKU from the sample, fewer sites are left to bind TSKU-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TSKU concentration in each sample is interpolated from this standard curve.<br><br>Intended Uses: This TSKU ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse TSKU.
