Application Notes
IP: 5mg (will immunoprecipitated CREB from 500mg of HeLa nuclear extract).
WB: 0.5-2mg/ml detects CREB from a HeLa nuclear lysate. Representative blot from a previous lot: HeLa nuclear lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-CREB (0.5ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.
Applications are based on unconjugated antibody.
Immunogen
KLH-conjugated, synthetic peptide (C-SGAENQQSGDAAVTEAENQQ) corresponding to amino acids 5-24 of human CREB. The immunizing sequence is identical in pig, shares 19 of 20 residues with mouse and bovine and 18 of 20 residues with rat.

HeLa nuclear extract. Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels.

Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

NCBI Description

This gene encodes a transcription factor that is a member of the leucine zipper family of DNA binding proteins. This protein binds as a homodimer to the cAMP-responsive element, an octameric palindrome. The protein is phosphorylated by several protein kinases, and induces transcription of genes in response to hormonal stimulation of the cAMP pathway. Alternate splicing of this gene results in several transcript variants encoding different isoforms. [provided by RefSeq, Mar 2016]
