Preparation and Storage

Corin, is a glycosylated protease on the cell surface as a single pass type II membrane protein. It is located on trial and brain natriurectic peptides (ANP and NNP) expressing cell surfaces. It may occur in both zymogen and catlytically active forms. The multi-domain transmembrane serine protease corin cleaves pro-atrial natriuretic peptide (pro-ANP) in vitro to generate an active hormone, ANP at R123-S124. Corin is higher expressed on cardiomyocytes. It may also contribute to the regulation of the natriuretic peptide system in vivo, and might be an attractive target for treatment of cardiovascular diseases.

NCBI Description

This gene encodes a member of the type II transmembrane serine protease class of the trypsin superfamily. Members of this family are composed of multiple structurally distinct domains. The encoded protein converts pro-atrial natriuretic peptide to biologically active atrial natriuretic peptide, a cardiac hormone that regulates blood volume and pressure. This protein may also function as a pro-brain-type natriuretic peptide convertase. [provided by RefSeq]

Function: Serine-type endopeptidase involved in atrial natriuretic peptide hormone processing. Converts through proteolytic cleavage the non-functional propeptide NPPA/ANP into the active hormone which promotes natriuresis, diuresis, and vasodilatation. May also process pro-NPPB the B-type natriuretic peptide. Ref.5

Enzyme regulation: Inhibited in a dose-dependent manner by non-specific trypsin-like serine protease inhibitors including benzamidine.

Subcellular location: Cell membrane; Single-pass type II membrane protein.

Tissue specificity: Highly expressed in heart. Expressed in heart myocytes. Ref.1

Post-translational modification: N-glycosylated; required for processing and activation

By similarity. Ref.7Activated through proteolytic processing by a trypsin-like protease; cleaved into a N-terminal propeptide and an activated corin protease fragment. Ref.7A disulfide bond links the N-terminal propeptide and the activated corin protease fragment.
