ICC: 1:100-1:500
IHC: 1:50-1:200
ELISA(peptide): 1:20,000-1:40,000
Western Blot (WB)
(Western blot analysis of extracts from B16F10 cells, using CLCA1 Antibody. The lane on the left was treated with blocking peptide.)
Immunohistochemistry (IHC)-Paraffin
(MBS9612260 at 1/100 staining Human colorectal cancer by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the primary antibody at 4°C overnight. An HRP conjugated anti-Rabbit antibody was used as the secondary antibody.)
Immunofluorescence (IF)/Immunocytochemistry (ICC)
(MBS9612260 staining Hela cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(MBS9612260 1:200) and mouse anti-beta tubulin Ab(T0023 1:200) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI(blue).)
Post Translational Modifications: Glycosylated. The 125-kDa product is autoproteolytically processed by the metalloprotease domain and yields to two cell-surface-associated subunits, a 90-kDa protein and a group of 37- to 41-kDa proteins. The cleavage is necessary for calcium-activated chloride channel (CaCC) activation activity.
Subcellular Location: Secreted>Extracellular space. Cell membrane>Peripheral membrane protein>Extracellular side. Note: Protein that remains attached to the plasma membrane appeared to be predominantly localized to microvilli.
Tissue Specificity: Highly expressed in small intestine and colon namely in intestinal basal crypt epithelia and goblet cells, and appendix. Weakly expressed in uterus, testis and kidney. Expressed in the airways epithelium of both asthmatic and healthy patients. Expressed in the bronchial epithelium, especially in mucus-producing goblet cells. Expressed in normal turbinate mucosa and nasal polyp. Expressed in.
Similarity: The metalloprotease region is responsible for autoproteolytic processing. It can also cross-cleave other CLCA substrates.Belongs to the CLCR family.
NCBI and Uniprot Product Information
Predicted Molecular Weight: (Calculated)100kDa.
NCBI Description
This gene encodes a member of the calcium sensitive chloride conductance protein family. To date, all members of this gene family map to the same region on chromosome 1p31-p22 and share a high degree of homology in size, sequence, and predicted structure, but differ significantly in their tissue distributions. The encoded protein is expressed as a precursor protein that is processed into two cell-surface-associated subunits, although the site at which the precursor is cleaved has not been precisely determined. The encoded protein may be involved in mediating calcium-activated chloride conductance in the intestine. [provided by RefSeq, Jul 2008]
Uniprot Description
CLCA1: May be involved in mediating calcium-activated chloride conductance. May play critical roles in goblet cell metaplasia, mucus hypersecretion, cystic fibrosis and AHR. May be involved in the regulation of mucus production and/or secretion by goblet cells. Involved in the regulation of tissue inflammation in the innate immune response. May play a role as a tumor suppressor. Induces MUC5AC. Belongs to the CLCR family.

Product Notes
The CLCA1 clca1 (Catalog #AAA9612260) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The CLCA1 Antibody reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's CLCA1 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). WB: 1:500-1:2000 IF: 1:100-1:500 ICC: 1:100-1:500 IHC: 1:50-1:200 ELISA(peptide): 1:20,000-1:40,000. Researchers should empirically determine the suitability of the CLCA1 clca1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CLCA1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
