Western Blot (WB) (Figure 1. Western blot analysis of CHX10 using anti-CHX10 antibody (MBS1750894). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: mouse eye ball tissue lysate. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CHX10 antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for CHX10 at approximately 43KD. The expected band size for CHX10 is at 39KD. )

E Coli-derived human CHX10 recombinant protein (Position: A249-A361). Human CHX10 shares 90.3% amino acid (aa) sequence identity with mouse CHX10.
Subcellular Localization
Nucleus.
Tissue Specificity
Abundantly expressed in retinal neuroblasts during eye development and in the inner nuclear layer of the adult retina. Within this layer, expression is stronger in the outer margin where bipolar cells predominate.
Reconstitution
Add 0.2ml of distilled water will yield a concentration of 500ug/ml.
Contents
Each vial contains 5mg BSA, 0.9mg NaCl, 0.2mg Na2HPO4, 0.05mg NaN3.
Preparation and Storage
Store at -20 degree C for one year. After reconstitution, at 4 degree C for one month. It can also be aliquotted and stored frozen at -20 degree C for a longer time. Avoid repeated freezing and thawing.

Description: Visual system homeobox 2 is a protein that in humans is encoded by the VSX2 gene. This gene encodes a homeobox protein originally described as a retina-specific transcription factor. Mutations in this gene are associated with microphthalmia, cataracts and iris abnormalities. In situ hybridization to human fetal retinal sections detected CHX10 expression in retinal neuroblasts at all stages examined. Human CHX10 is expressed in progenitor cells of the developing neuroretina and in the inner nuclear layer of the mature retina.
