Specificity
This item detects CD279, a co-stimulatory molecule also known as programmed cell death-1 (PD-1). CD279 is a single-pass type 1 membrane protein belonging to the CD28 family, and functions mainly as a negative regulator of T-cell activation. CD279 has two specific ligands; CD274 (PD-L1) and CD273 (PD-L2), and their interaction is key in the balance between stimulatory and inhibitory signals needed for effective immune responses to microbes and self-tolerance. CD279 is inducibly expressed by T-cells, B-cells, NK-T-cells and monocytes upon activation.

Loss of CD279 function has been associated with a number of autoimmune diseases, including rheumatoid arthritis, type I diabetes and ankylosing spondylitis. Recent studies suggest that CD279 could be targeted therapeutically in the treatment of HIV infection to reduce T-cell exhaustion.

NCBI Description

This gene encodes a cell surface membrane protein of the immunoglobulin superfamily. This protein is expressed in pro-B-cells and is thought to play a role in their differentiation. In mice, expression of this gene is induced in the thymus when anti-CD3 antibodies are injected and large numbers of thymocytes undergo apoptosis. Mice deficient for this gene bred on a BALB/c background developed dilated cardiomyopathy and died from congestive heart failure. These studies suggest that this gene product may also be important in T cell function and contribute to the prevention of autoimmune diseases. [provided by RefSeq, Jul 2008]

Product Notes

The CD279 pdcd1 (Catalog #AAA224115) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI HUMAN CD279 (C-TERMINAL) reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's CD279 can be used in a range of immunoassay formats including, but not limited to, Immunohistology Paraffin*, Western Blot (WB). Western Blot: This item detects a band of approximately 45kDa in THP-1 cell lysate Immunohistology - Paraffin: Maximum Dilution: 2.5ug/ml; Application Note: This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose. Western Blotting: Minimum Dilution: 0.5; Maximum Dilution: 1.0ug/ml. Researchers should empirically determine the suitability of the CD279 pdcd1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD279, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
