Western Blot (WB)
(Figure 1. Western blot analysis of CD11a using anti-CD11a antibody (MBS178688).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.lane 1: JURKAT cell lysates, lane 2: CEM whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CD11a antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for CD11a at approximately 150KD. The expected band size for CD11a is at 129KD. )
Flow Cytometry (FC/FACS)
(Figure 2. Flow Cytometry analysis of U937 cells using anti-CD11a antibody (MBS178688).Overlay histogram showing U937 cells stained with MBS178688 (Blue line). The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-CD11a Antibody (MBS178688,1ug/1x10^6 cells) for 30 min at 20 degree C. DyLight 488 conjugated goat anti-rabbit IgG (5-10ug/1x10^6 cells) was used as secondary antibody for 30 minutes at 20 degree C. Isotype control antibody (Green line) was rabbit IgG (1ug/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control.")
Background: CD11a, also known as ITGAL or LFA-1, is a human gene which functions in the immune system. It is mapped to 16p11.2. CD11a is involved in cellular adhesion and costimulatory signaling. It is the target of the drug efalizumab. CD11a encodes the integrin alpha L chain. Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. This I-domain containing alpha integrin combines with the beta 2 chain (ITGB2) to form the integrin lymphocyte function-associated antigen-1 (LFA-1), which is expressed on all leukocytes. CD11a plays a central role in leukocyte intercellular adhesion through interactions with its ligands, ICAMs 1-3 (intercellular adhesion molecules 1 through 3), and also functions in lymphocyte costimulatory signaling. It is one of the two components, along with CD18, which form lymphocyte function-associated antigen-1.

NCBI Description
ITGAL encodes the integrin alpha L chain. LFA-1 plays a central role in leukocyte intercellular adhesion through interactions with its ligands, ICAMs 1-3 (intercellular adhesion molecules 1 through 3), and also functions in lymphocyte costimulatory signaling. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
Uniprot Description
ITGAL: Integrin alpha-L/beta-2 is a receptor for ICAM1, ICAM2, ICAM3 and ICAM4. It is involved in a variety of immune phenomena including leukocyte-endothelial cell interaction, cytotoxic T-cell mediated killing, and antibody dependent killing by granulocytes and monocytes. Belongs to the integrin alpha chain family. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes
The ITGAL itgal (Catalog #AAA178688) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-CD11a Antibody reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's CD11a can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Western Blot: 0.1-0.5ug/ml. Researchers should empirically determine the suitability of the ITGAL itgal for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD11a, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
