Cathepsin D

Goat polyclonal antibody to Cathepsin D. Cathepsin D is a lysosomal aspartic protease of the pepsin family. Acid protease active in intracellular protein breakdown. It is composed of a dimer of disulfide-linked heavy and light chains, both produced from a single protein precursor. It is involved in the pathogenesis of several diseases such as breast cancer and possibly Alzheimer disease.

NCBI Description

This gene encodes a lysosomal aspartyl protease composed of a dimer of disulfide-linked heavy and light chains, both produced from a single protein precursor. This proteinase, which is a member of the peptidase C1 family, has a specificity similar to but narrower than that of pepsin A. Transcription of this gene is initiated from several sites, including one which is a start site for an estrogen-regulated transcript. Mutations in this gene are involved in the pathogenesis of several diseases, including breast cancer and possibly Alzheimer disease. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Acid protease active in intracellular protein breakdown. Involved in the pathogenesis of several diseases such as breast cancer and possibly Alzheimer disease.

Catalytic activity: Specificity similar to, but narrower than, that of pepsin A. Does not cleave the 4-Gln-|-His-5 bond in B chain of insulin.

Subunit structure: Consists of a light chain and a heavy chain.

Subcellular location: Lysosome. Melanosome. Secreted › extracellular space. Note: Identified by mass spectrometry in melanosome fractions from stage I to stage IV. In aortic samples, detected as an extracellular protein loosely bound to the matrix (Ref.17). Ref.10 Ref.14 Ref.17

Tissue specificity: Expressed in the aorta extrcellular space (at protein level). Ref.17

Post-translational modification: N- and O-glycosylated. Ref.11 Ref.19

Polymorphism: The Val-58 allele is significantly overrepresented in demented patients (11.8%) compared with non-demented controls (4.9%). Carriers of the Val-58 allele have a 3.1-fold increased risk for developing AD than non-carriers.

Involvement in disease: Ceroid lipofuscinosis, neuronal, 10 (CLN10) [MIM:610127]: A form of neuronal ceroid lipofuscinosis with onset at birth or early childhood. Neuronal ceroid lipofuscinoses are progressive neurodegenerative, lysosomal storage diseases characterized by intracellular accumulation of autofluorescent liposomal material, and clinically by seizures, dementia, visual loss, and/or cerebral atrophy.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.13 Ref.23 Ref.24

Sequence similarities: Belongs to the peptidase A1 family.

Product Notes

The CTSD ctsd (Catalog #AAA448021) is an Antibody produced from Goat and is intended for research purposes only. The product is available for immediate purchase. The Cathepsin D Polyclonal Antibody reacts with Reacts against human, rat, mouse, canine and monkey proteins. and may cross-react with other species as described in the data sheet. AAA Biotech's Cathepsin D can be used in a range of immunoassay formats including, but not limited to, Western Blot, Immunofluorescence, Immunohistochemistry (paraffin), Immunohistochemistry (frozen), IEM. Western Blot: 1:250-1:1,000
Immunofluorescence 1:50-1:200
Immunohistochemistry (paraffin) 1:50-200
Immunohistochemistry (frozen) 1:50-1:200
IEM 1:50-1:200. Researchers should empirically determine the suitability of the CTSD ctsd for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Cathepsin D, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
