• Application Notes: Suitable for use in Western Blot.; Dilution: Western Blot: 1:1000. Incubate membrane with diluted antibody in 5% w/v BSA, 1X TBS, 0.1% Tween-20 at 4 degree C with gentle shaking, overnight.
• Immunogen: Synthetic peptide corresponding to a region surrounding residue Ser37 of human B-catenin (blue carrier-coupled).
• Preparation and Storage: May be stored at 4 degree C for short-term only. For long-term storage, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

B-catenin is a key downstream effector in the Wnt signaling pathway (1). It is implicated in two major biological processes in vertebrates: early embryonic development (2) and tumorigenesis (3). CK1 phosphorylates B-catenin on Ser45. This phosphorylation event primes B-catenin for subsequent phosphorylation by GSK-3 (4-6). GSK-3B destabilizes B-catenin by phosphorylating it at Ser33, Ser37 and Thr41 (7). Mutations in these phosphorylation sites, which result in the stabilization of B-catenin protein levels, have been found in many tumor cell lines (8).
