Immunohistochemistry (IHC)

(Immunohistochemical analysis of c-Myb staining in human tonsil formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.)

Immunofluorescence (IF)

(Immunofluorescent analysis of c-Myb staining in HeLa cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.)

NCBI Description

This gene encodes a transcription factor that is a member of the MYB family of transcription factor genes. The protein contains three domains, an N-terminal DNA-binding domain, a central transcriptional activation domain and a C-terminal domain involved in transcriptional repression. This protein plays an essential role in the regulation of hematopoiesis and may play a role in tumorigenesis. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jun 2009]

Uniprot Description

Myb: Transcriptional activator; DNA-binding protein that specifically recognize the sequence 5'-YAAC[GT]G-3'. Plays an important role in the control of proliferation and differentiation of hematopoietic progenitor cells. 6 isoforms of the human protein are produced by alternative splicing.

Protein type: DNA-binding; Oncoprotein; Transcription factor

Chromosomal Location of Human Ortholog: 6q22-q23

Cellular Component: nuclear matrix; cytosol

Molecular Function: protein binding; chromatin binding

Biological Process: positive regulation of histone H3-K9 methylation; transcription, DNA-dependent; thymus development; in utero embryonic development; positive regulation of transcription, DNA-dependent; homeostasis of number of cells; positive regulation of histone H3-K4 methylation; negative regulation of transcription from RNA polymerase II promoter; chromatin remodeling; positive regulation of T-helper cell differentiation; regulation of transcription, DNA-dependent; calcium ion transport; B cell differentiation; positive regulation of transcription from RNA polymerase II promoter; negative regulation of transcription, DNA-dependent; blood coagulation; embryonic gut development; G1/S transition of mitotic cell cycle

Product Notes

The MYB myb (Catalog #AAA8240689) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-c-Myb Antibody reacts with Human, Mouse, Rat, Bovine and may cross-react with other species as described in the data sheet. AAA Biotech's c-Myb can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB),Immunohistochemistry (IHC), Immunofluorescence (IF), Immunocytochemistry (ICC). WB (1/500 - 1/1000), IHC (1/100 - 1/200), IF (1/100 - 1/500), ICC (1/100 - 1/500). Researchers should empirically determine the suitability of the MYB myb for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "c-Myb, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
