Function
Non-receptor tyrosine kinase which mediatessignal transduction downstream of a varietyof transmembrane receptors includingclassical immunoreceptors like the B-cellreceptor (BCR). Regulates several biological processes including innate and adaptive immunity, cell adhesion, osteoclastmaturation, platelet activation and vasculardevelopment. Assembles into signalingcomplexes with activated receptors at theplasma membrane via interaction between itsSH2 domains and the receptor tyrosine-phosphorylated ITAM domains. Theassociation with the receptor can also beindirect and mediated by adapter proteins containing ITAM or partial hemITAM domains.The phosphorylation of the ITAM domains isgenerally mediated by SRC subfamily kinases upon engagement of the receptor. Morerarely signal transduction via SYK could be ITAM-independent. Direct down stream effectors phosphorylated by SYK include VAV1, PLCG1, PI-3-kinase, LCP2 and BLNK. Initially identified as essential in B-cellreceptor (BCR) signaling, it is necessary forthe maturation of B-cells most probably at the pro-B to pre-B transition. Activated upon BCR engagement, it phosphorylates and activates BLNK an adapter linking the activated BCR to downstream signaling adapters and effectors. It also phosphorylates and activates PLCG1 and the PKC signaling pathway. It also phosphorylates BTK and regulates its activity in B-cell antigenreceptor (BCR)-coupled signaling. In addition to its function downstream of BCR plays alsoa role in T- cell receptor signaling. Plays alsoa crucial role in the innate immune response to fungal, bacterial and viral pathogens. It isfor instance activated by the membranelectin CLEC7A. Upon stimulation by fungalproteins, CLEC7A together with SYK activates immune cells inducing the production of ROS. Also activates the inflammasome and NF-kappa-B-mediated transcription of chemokines and cytokines in presence ofpathogens. Regulates neutrophildegranulation and phagocytosis throughactivation of the MAPK signaling cascade (Bysimilarity). Required for the stimulation ofneutrophil phagocytosis by IL15 (PubMed:15123770). Alsomediates the activation of dendritic cells bycell necrosis stimuli. Also involved in mastcells activation. Involved in interleukin-3/IL3-mediated signaling pathway in basophils (Bysimilarity). Also functions downstream ofreceptors mediating cell adhesion. Relays forinstance, integrin-mediated neutrophils andmacrophages activation and P-selectinreceptor/SELPG-mediated recruitment ofleukocytes to inflammatory loci. Plays also arole in non-immune processes. It is for instance involved in vascular development where it may regulate blood and lymphaticvascular separation. It is also required forosteoclast development and function.Functions in the activation of platelets bycollagen, mediating PLCG2 phosphorylation and activation. May be coupled to thecollagen receptor by the ITAMdomain-containing FCER1G. Also activated bythe membrane lectin CLEC1B that is required for activation of platelets byPDPN/podoplanin. Involved in platelet adhesion being activated by ITGB3 engagedby fibrinogen. Together with CEACAM20,enhances production of the cytokineCXCL8/IL-8 via the NFKB pathway and maythus have a role in the intestinal immuneresponse (By similarity).

Preparation and Storage

Syk is a positive effector of BCR-stimulated responses. This protein couples the B-cell antigen receptor (BCR) to the mobilization of calcium ion either through a phosphoinositide 3-kinase-dependent pathway, when not phosphorylated on tyrosines of the linker region, or through a phospholipase C-gamma-dependent pathway, when phosphorylated on Tyr-342 and Tyr-346. Thus the differential phosphorylation of Syk can determine the pathway by which BCR is coupled to the regulation of intracellular calcium ion.

NCBI Description

This gene encodes a member of the family of non-receptor type Tyr protein kinases. This protein is widely expressed in hematopoietic cells and is involved in coupling activated immunoreceptors to downstream signaling events that mediate diverse cellular responses, including proliferation, differentiation, and phagocytosis. It is thought to be a modulator of epithelial cell growth and a potential tumour suppressor in human breast carcinomas. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Mar 2010]

Syk: a cytoplasmic tyrosine kinase of the SYK family containing two SH2 domains. Plays a central role in the B cell receptor (BCR) response. An upstream activator of the PI3K, PLCgamma2, and Rac/cdc42 pathways in the BCR response. Required for the sequential events of Fc gamma IIa receptor-mediated phagocytosis. Expression highest in murine spleen, heart, mammary gland and thymus. Two splice variant isoforms have been described.
