This gene encodes a protein that regulates autophagy, a catabolic process of degradation induced by starvation. The encoded protein is a component of the phosphatidylinositol-3-kinase (PI3K) complex which mediates vesicle-trafficking processes. This protein is thought to play a role in multiple cellular processes, including tumorigenesis, neurodegeneration and apoptosis. Alternative splicing results in multiple transcript variants. beclin 1: an essential autophagy protein that has been linked to multiple processes including tumor suppression, protection against some cardiac and neurological degenerative diseases, and lifespan extension. A ubiquitous protein of the beclin family. Is part of the class III PI3K complex that induces autophagy. Expressed at elevated levels in dendrites and cell bodies of cerebellar Purkinje cells. Interacts with the anti-apoptotic protein Bcl-2, which inhibits autophagy when it is present in the endoplasmic reticulum. The dissociation of ATG6 from Bcl-2 is essential for its autophagic activity. Interacts with PITC and GluR-delta2. Probably forms a complex with AMBRA1 and PIK3C3. Plays a role in antiviral host defense. Protects against infection by a neurovirulent strain of Sindbis virus.
