Related Product Information for anti-ATG4B antibody
Macroautophagy is the major inducible pathway for the general turnover of cytoplasmic constituents in eukaryotic cells, it is also responsible for the degradation of active cytoplasmic enzymes and organelles during nutrient starvation. Macroautophagy involves the formation of double-membrane bound autophagosomes which enclose the cytoplasmic constituent targeted for degradation in a membrane bound structure, which then fuse with the lysosome (or vacuole) releasing a single-membrane bound autophagic bodies which are then degraded within the lysosome (or vacuole). APG4, a cysteine protease required for autophagy, cleaves the C-terminal part of MAP1LC3 to form the activated molecule LC3-I. LC3-I is subsequently conjugated with phosphatidylethanolamine at the C-terminal glycine to form LC3-II, a marker for autophagy via its capacity to bind to autophagosomes.

NCBI Description

Autophagy is the process by which endogenous proteins and damaged organelles are destroyed intracellularly. Autophagy is postulated to be essential for cell homeostasis and cell remodeling during differentiation, metamorphosis, non-apoptotic cell death, and aging. Reduced levels of autophagy have been described in some malignant tumors, and a role for autophagy in controlling the unregulated cell growth linked to cancer has been proposed. This gene encodes a member of the autophagin protein family. The encoded protein is also designated as a member of the C-54 family of cysteine proteases. Alternate transcriptional splice variants, encoding different isoforms, have been characterized. [provided by RefSeq, Jul 2008]

Uniprot Description

ATG4B: Cysteine protease required for autophagy, which cleaves the C-terminal part of either MAP1LC3, GABARAPL2 or GABARAP, allowing the liberation of form I. A subpopulation of form I is subsequently converted to a smaller form (form II). Form II, with a revealed C-terminal glycine, is considered to be the phosphatidylethanolamine (PE)-conjugated form, and has the capacity for the binding to autophagosomes. Mainly expressed in the skeletal muscle, followed by brain, heart, liver and pancreas. Inhibited by N-ethylmaleimide. Belongs to the peptidase C54 family. 5 isoforms of the human protein are produced by alternative splicing.

Protein type: Autophagy; Ubiquitin conjugating system; EC 3.4.22.-; Protease

Chromosomal Location of Human Ortholog: 2q37.3

Cellular Component: cytoplasm; cytosol

Molecular Function: protein binding; cysteine-type endopeptidase activity; endopeptidase activity; cysteine-type peptidase activity

Product Notes

The ATG4B atg4b (Catalog #AAA9205832) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 258-284. The ATG4B Antibody reacts with Human (Predicted Reactivity: Bovine, Chicken, Zebrafish, Mouse) and may cross-react with other species as described in the data sheet. AAA Biotech's ATG4B can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), ELISA (EIA), Immunohistochemistry (IHC). WB~~1:1000. Researchers should empirically determine the suitability of the ATG4B atg4b for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ATG4B, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
