Polyclonal Antibody to detect ApoER2 in human samples

Background: ApoER2 (apolipoprotein E receptor 2), also designated LRP8, is a member of the LDL receptor gene family, which includes LDL receptor, LRP, megalin, VLDLR and ApoER2. The LDL receptor family is characterized by a cluster of cysteine-rich class A repeats, epidermal growth factor (EGF)-like repeats, YWTD repeats and an O-linked sugar domain. ApoER2 is expressed in brain and placenta and has several splice variants. ApoER2 is thought to mediate the interaction of extracellular Reelin and cytosolic mDab1 (mammalian disabled protein), which activates a tyrosine kinase. This pathway regulates the migration of neurons along the radial glial fiber network during brain development.

NCBI Description

This gene encodes a member of the low density lipoprotein receptor (LDLR) family. Low density lipoprotein receptors are cell surface proteins that play roles in both signal transduction and receptor-mediated endocytosis of specific ligands for lysosomal degradation. The encoded protein plays a critical role in the migration of neurons during development by mediating Reelin signaling, and also functions as a receptor for the cholesterol transport protein apolipoprotein E. Expression of this gene may be a marker for major depressive disorder. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. Uniprot Description

LRP8: Cell surface receptor for Reelin (RELN) and apolipoprotein E (apoE)-containing ligands. LRP8 participates in transmitting the extracellular Reelin signal to intracellular signaling processes, by binding to DAB1 on its cytoplasmic tail. Reelin acts via both the VLDL receptor (VLDLR) and LRP8 to regulate DAB1 tyrosine phosphorylation and microtubule function in neurons. LRP8 has higher affinity for Reelin than VLDLR. LRP8 is thus a key component of the Reelin pathway which governs neuronal layering of the forebrain during embryonic brain development. Binds the endoplasmic reticulum resident receptor-associated protein (RAP). Binds dimers of beta 2-glycoprotein I and may be involved in the suppression of platelet aggregation in the vasculature. Highly expressed in the initial segment of the epididymis, where it affects the functional expression of clusterin and phospholipid hydroperoxide glutathione peroxidase (PHGPx), two proteins required for sperm maturation. May also function as an endocytic receptor. Defects in LRP8 are a cause of myocardial infarction type 1 (MCI1). A condition defined by the irreversible necrosis of heart muscle secondary to prolonged ischemia. Belongs to the LDLR family. 5 isoforms of the human protein are produced by alternative splicing.

Product Notes

The LRP8 lrp8 (Catalog #AAA840258) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The ApoER2 (LRP8) Antibody (CT) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's ApoER2 (LRP8) can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC). Western blot: ~1:1000, IHC: ~1:50-1:100. Researchers should empirically determine the suitability of the LRP8 lrp8 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ApoER2 (LRP8), Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
