Immunofluorescence (IF)
(Fluorescent confocal image of C2C12 cell stained with AKT2 Antibody (S474).C2C12 cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with AKT2 primary antibody (1:25, 1 h at 37 degree C). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:400, 50 min at 37 degree C).Cytoplasmic actin was counterstained with Alexa Fluor 555 (red) conjugated Phalloidin (7units/ml, 1 h at 37 degree C). Nuclei were counterstained with DAPI (blue) (10 ug/ml, 10 min).AKT2 immunoreactivity is localized to Cytoplasm significantly.)

Akt2: an AGC kinase. Plays critical roles in glucose metabolism and the development or maintenance of proper adipose tissue and islet mass for which other Akt/PKB isoforms are unable to fully compensate. Amplified and overexpressed in human ovarian carcinoma cell lines and amplified in some primary ovarian and pancreatic tumors. Antisense blocks invasiveness in xenografts. Expressed in several insulin-responsive tissues, and one case of Type II diabetes has been associated with a likely LOF point mutation. Mouse mutants have defects in insulin response.
