NCBI and Uniprot Product Information
NCBI Description
Encodes AKT2, a photosynthate- and light-dependent inward rectifying potassium channel with unique gating properties that are regulated by phosphorylation. Expressed in guard cell protoplasts and in the phloem and xylem of aerial portions of the plant. The channel can coassemble with another K+ channel, KAT1, in vitro. In guard cells, AKT2/3 is responsible for the Ca2+ sensitivity of the K+ uptake channel. In the phloem, it regulates the sucrose/H+ symporters via the phloem potential. AKT2 belongs to the Shaker family K+ channels which include the following groups based on phylogenetic analysis (FEBS Letters (2007) 581: 2357): I (inward rectifying channel): AKT1 (AT2G26650), AKT5 (AT4G32500) and SPIK (also known as AKT6, AT2G25600); II (inward rectifying channel): KAT1 (AT5G46240) and KAT2 (AT4G18290); III (weakly inward rectifying channel): AKT2 (AT4G22200); IV (regulatory subunit involved in inwardly rectifying conductance formation): KAT3 (also known as AtKC1, AT4G32650); V (outward rectifying channel): SKOR (AT3G02850) and GORK (AT5G37500).
Uniprot Description
Function: Highly selective and weak inward-rectifying potassium channel. Plays a role in both loading and unloading potassium into/from the phloem sap. Seems to control sugar loading into phloem via a voltage-dependent process. Blocked by physiological concentrations of external calcium and by external acidification. May interact with the cytoskeleton or with regulatory proteins. Dephosphorylation by PP2CA not only leads to the inhibition of potassium currents but also to an increase of the voltage-dependence of the channel. Ref.11 Ref.14
Subunit structure: The potassium channel is probably composed of a homo- or heterotetrameric complex of pore-forming subunits. Interacts with the phosphatase PPC2A. May interact with AKT1, KAT1 and KAT3. Ref.7 Ref.9 Ref.12 Ref.14
Subcellular location: Membrane; Multi-pass membrane protein.
Tissue specificity: Expressed mainly in the phloem tissues throughout the plant but also, at a lower level, in leaf epiderm, mesophyll and guard cells. Ref.1 Ref.6 Ref.8 Ref.12
Induction: In shoots, strongly induced by abscisic acid (ABA) treatment and reduced after NaCl treatment or potassium starvation. Ref.14
Domain: The segment S4 is probably the voltage-sensor and is characterized by a series of positively charged amino acids. The pore-forming region H5 is enclosed by the transmembrane segments S5 and S6 in the Shaker-type (1P/6TM) and contains the GYGD signature motif which seems to be involved in potassium selectivity. The KHA domain (rich in hydrophobic and acidic residues) present in the C-terminal part is likely to be important for tetramerization.
Post-translational modification: Dephosphorylated by PP2CA. Ref.12
Sequence similarities: Belongs to the potassium channel family. Plant (TC 1.A.1.4) subfamily. [View classification]Contains 5 ANK repeats.Contains 1 cyclic nucleotide-binding domain.Contains 1 KHA domain.
Sequence caution: The sequence AAA96153.1 differs from that shown. Reason: Erroneous initiation. The sequence AAA96154.1 differs from that shown. Reason: Erroneous initiation. The sequence CAA16770.1 differs from that shown. Reason: Erroneous initiation. The sequence CAB79175.1 differs from that shown. Reason: Erroneous initiation.

Product Notes
The KT2/3 akt2 (Catalog #AAA622883) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Akt2,3 (AKT-2, Protein Kinase Akt2, Protein Kinase B beta, PKB beta, PKBBeta, PRKBB, Rac Protein Kinase beta, RAC-BETA, RAC-beta Serine/threonine Protein Kinase, RAC-PK-beta, v-AKT Murine Thymoma Viral Oncogene Homolog 2) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Akt2,3 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Suitable for use in Western Blot. Researchers should empirically determine the suitability of the KT2/3 akt2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Akt2,3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
