Antisera to AIF were raised by repeated immunisation of rabbit with highly purified antigen. Purified IgG was prepared from whole serum by ion exchange chromatography.

Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. This product should be stored undiluted. Storage in frost free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the protein. Should this product contain a precipitate we recommend microcentrifugation before use.

Rabbit anti Human AIF antibody recognizes human Apoptosis-inducing factor 1, mitochondrial, also known as Programmed cell death protein 8 or apoptosis inducing factor (AIF). AIF is a 613 amino acid member of the FAD-dependent oxidoreductase family.Rabbit anti Human AIF antibody reacts with an internal region of AIF (within the region aa 517-531. AIF is widely expressed in human tissues and localizes in the mitochondria. AIF translocates to the nucleus upon induction of apoptosis. Multiple isoforms of AIF are generated by alternative splicing. Rabbit anti Human AIF antibody recognizes isoforms 1, 2,3 and 5. It is not expected to recognize either isoforms 4 or 6 as they lack the immunogen sequence. Mutations in the AIFM1 can result in various disease conditions including COXPD6 and Cowchock syndrome.

NCBI Description

This gene encodes a flavoprotein essential for nuclear disassembly in apoptotic cells, and it is found in the mitochondrial intermembrane space in healthy cells. Induction of apoptosis results in the translocation of this protein to the nucleus where it affects chromosome condensation and fragmentation. In addition, this gene product induces mitochondria to release the apoptogenic proteins cytochrome c and caspase-9. Mutations in this gene cause combined oxidative phosphorylation deficiency 6, which results in a severe mitochondrial encephalomyopathy. Alternative splicing results in multiple transcript variants. A related pseudogene has been identified on chromosome 10. [provided by RefSeq, May 2010]

PDCD8: Probable oxidoreductase that has a dual role in controlling cellular life and death; during apoptosis, it is translocated from the mitochondria to the nucleus to function as a proapoptotic factor in a caspase-independent pathway, while in normal mitochondria, it functions as an antiapoptotic factor via its oxidoreductase activity. The soluble form (AIFsol) found in the nucleus induces 'parthanatos' i.e., caspase-independent fragmentation of chromosomal DNA. Interacts with EIF3G,and thereby inhibits the EIF3 machinery and protein synthesis, and activates casapse-7 to amplify apoptosis. Plays a critical role in caspase- independent, pyknotic cell death in hydrogen peroxide-exposed cells. Binds to DNA in a sequence-independent manner. Interacts with XIAP/BIRC4. Interacts (via N-terminus) with EIF3G (via C-terminus). Belongs to the FAD-dependent oxidoreductase family. 4 isoforms of the human protein are produced by alternative splicing.
