Goat AGRP/Agouti-related protein Polyclonal Antibody | anti-AGRP antibody
Goat anti-AGRP/Agouti-related protein Antibody
Immunohistochemistry: Frozen section of Human Hypothalamus shows staining of a group of neuronal cell bodies in the human infundibular (arcuate) nucleus and their varicose axons. Recommended concentration, 0.05-0.1ug/ml.
Western Blot: Preliminary experiments gave bands at approx 45kDa and 24kDa in Human Brain lysates after 0.1ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the bands we observe given the calculated size of 14.4kDa according to NP_001129.1and NP_015531.1. Both detected bands were successfully blocked by incubation with the immunizing peptide (and BLAST results with the immunizing peptide sequence did not identify any other proteins to explain the additional bands).
Immunohistochemistry (IHC)
((0. 05ug/ml) staining of PFA-perfused cryosection of Human Hypothalamus. Antigen retrieval with citrate buffer pH 6 at 80C for 30min, HRP-staining with Ni-DAB after Biotin-SP-antigoat amplification. Data obtained by Prof. Erik Hrabovszky, Inst, Exp, Med. , Budapest, Hungary.)
NCBI and Uniprot Product Information
NCBI Description
This gene encodes an antagonist of the melanocortin-3 and melanocortin-4 receptor. It appears to regulate hypothalamic control of feeding behavior via melanocortin receptor and/or intracellular calcium regulation, and thus plays a role in weight homeostasis. Mutations in this gene have been associated with late on-set obesity. [provided by RefSeq, Dec 2009]
Uniprot Description
AGRP: Plays a role in weight homeostasis. Involved in the control of feeding behavior through the central melanocortin system. Acts as alpha melanocyte-stimulating hormone antagonist by inhibiting cAMP production mediated by stimulation of melanocortin receptors within the hypothalamus and adrenal gland. Has very low activity with MC5R. Is an inverse agonist for MC3R and MC4R being able to suppress their constitutive activity. It promotes MC3R and MC4R endocytosis in an arrestin-dependent manner. Genetic variations in AGRP may be a cause of obesity (OBESITY). It is a condition characterized by an increase of body weight beyond the limitation of skeletal and physical requirements, as the result of excessive accumulation of body fat.

Product Notes
The AGRP agrp (Catalog #AAA420442) is an Antibody produced from Goat and is intended for research purposes only. The product is available for immediate purchase. The Goat anti-AGRP/Agouti-related protein Antibody reacts with Tested: Human; Expected from sequence similarity: Human, Dog, Pig and may cross-react with other species as described in the data sheet. AAA Biotech's AGRP/Agouti-related protein can be used in a range of immunoassay formats including, but not limited to, Peptide ELISA (EIA), Immunohistochemistry (IHC). Peptide ELISA: Antibody detection limit dilution 1: 32000. Immunohistochemistry: Frozen section of Human Hypothalamus shows staining of a group of neuronal cell bodies in the human infundibular (arcuate) nucleus and their varicose axons. Both detected bands were successfully blocked by incubation with the immunizing peptide (and BLAST results with the immunizing peptide sequence did not identify any other proteins to explain the additional bands). Researchers should empirically determine the suitability of the AGRP agrp for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: CRKLGTAMNP CSRT. It is sometimes possible for the material contained within the vial of "AGRP/Agouti-related protein, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
