Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate PLXNA2 in samples. An antibody specific for PLXNA2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPLXNA2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PLXNA2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PLXNA2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

This gene encodes a member of the plexin-A family of semaphorin co-receptors. Semaphorins are a large family of secreted or membrane-bound proteins that mediate repulsive effects on axon pathfinding during nervous system development. A subset of semaphorins are recognized by plexin-A/neuropilin transmembrane receptor complexes, triggering a cellular signal transduction cascade that leads to axon repulsion. This plexin-A family member is thought to transduce signals from semaphorin-3A and -3C. [provided by RefSeq, Jul 2008]

PLXNA2: Coreceptor for SEMA3A and SEMA6A. Necessary for signaling by SEMA6A and class 3 semaphorins and subsequent remodeling of the cytoskeleton. Plays a role in axon guidance, invasive growth and cell migration. Class 3 semaphorins bind to a complex composed of a neuropilin and a plexin. The plexin modulates the affinity of the complex for specific semaphorins, and its cytoplasmic domain is required for the activation of down- stream signaling events in the cytoplasm. Homodimer. The PLXNA2 homodimer interacts with a SEMA6A homodimer, giving rise to a heterotetramer. Interacts directly with NRP1 and NRP2. Interacts with RND1. Detected in fetal brain. Belongs to the plexin family. 2 isoforms of the human protein are produced by alternative splicing.
