Principle of the assay: PKCz ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PKCz antibody and an PKCz-HRP conjugate. The assay sample and buffer are incubated together with PKCz-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PKCz concentration since PKCz from samples and PKCz-HRP conjugate compete for the anti-PKCz antibody binding site. Since the number of sites is limited, as more sites are occupied by PKCz from the sample, fewer sites are left to bind PKCz-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PKCz concentration in each sample is interpolated from this standard curve.

NCBI Description

Protein kinase C (PKC) zeta is a member of the PKC family of serine/threonine kinases which are involved in a variety of cellular processes such as proliferation, differentiation and secretion. Unlike the classical PKC isoenzymes which are calcium-dependent, PKC zeta exhibits a kinase activity which is independent of calcium and diacylglycerol but not of phosphatidylserine. Furthermore, it is insensitive to typical PKC inhibitors and cannot be activated by phorbol ester. Unlike the classical PKC isoenzymes, it has only a single zinc finger module. These structural and biochemical properties indicate that the zeta subspecies is related to, but distinct from other isoenzymes of PKC. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]
