Principle of the Assay: PKCi ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PKCi antibody and an PKCi-HRP conjugate. The assay sample and buffer are incubated together with PKCi-HRP conjugate in pre-coated plate for one hour.After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PKCi concentration since PKCi from samples and PKCi-HRP conjugate compete for the anti-PKCi antibody binding site. Since the number of sites is limited, as more sites are occupied by PKCi from the sample, fewer sites are left to bind PKCi-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PKCi concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the protein kinase C (PKC) family of serine/threonine protein kinases. The PKC family comprises at least eight members, which are differentially expressed and are involved in a wide variety of cellular processes. This protein kinase is calcium-independent and phospholipid-dependent. It is not activated by phorbolesters or diacylglycerol. This kinase can be recruited to vesicle tubular clusters (VTCs) by direct interaction with the small GTPase RAB2, where this kinase phosphorylates glyceraldehyde-3-phosphate dehydrogenase (GAPD/GAPDH) and plays a role in microtubule dynamics in the early secretory pathway. This kinase is found to be necessary for BCL-ABL-mediated resistance to drug-induced apoptosis and therefore protects leukemia cells against drug-induced apoptosis. There is a single exon pseudogene mapped on chromosome X. [provided by RefSeq, Jul 2008]
