Reactivity
Human
Specificity
This assay has high sensitivity and excellent specificity for detection of human PGK1. No significant cross-reactivity or interference between human PGK1 and analogues was observed.
Sequence Length
417
Samples
Serum, plasma, tissue homogenates, cell lysates.
Assay Type
Sandwich
Detection Range
28 pg/ml-1800 pg/ml.
Sensitivity
The minimum detectable dose of human PGK1 is typically less than 7 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
Intra-assay Precision
Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision
Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
Preparation and Storage
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for PGK1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PGK1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PGK1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PGK1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

The protein encoded by this gene is a glycolytic enzyme that catalyzes the conversion of 1,3-diphosphoglycerate to 3-phosphoglycerate. The encoded protein may also act as a cofactor for polymerase alpha. Additionally, this protein is secreted by tumor cells where it participates in angiogenesis by functioning to reduce disulfide bonds in the serine protease, plasmin, which consequently leads to the release of the tumor blood vessel inhibitor angiostatin. The encoded protein has been identified as a moonlighting protein based on its ability to perform mechanistically distinct functions. Deficiency of the enzyme is associated with a wide range of clinical phenotypes hemolytic anemia and neurological impairment. Pseudogenes of this gene have been defined on chromosomes 19, 21 and the X chromosome. [provided by RefSeq, Jan 2014]

Product Notes

The Human PGK1 pgk1 (Catalog #AAA941880) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA941880 ELISA Kit recognizes Human PGK1. It is sometimes possible for the material contained within the vial of "phosphoglycerate kinase 1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
