Principle of the Assay: PDXP ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PDXP antibody and an PDXP-HRP conjugate. The assay sample and buffer are incubated together with PDXP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PDXP concentration since PDXP from samples and PDXP-HRP conjugate compete for the anti-PDXP antibody binding site. Since the number of sites is limited, as more sites are occupied by PDXP from the sample, fewer sites are left to bind PDXP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PDXP concentration in each sample is interpolated from this standard curve.

NCBI Description

Pyridoxal 5-prime-phosphate (PLP) is the active form of vitamin B6 that acts as a coenzyme in maintaining biochemical homeostasis. The preferred degradation route from PLP to 4-pyridoxic acid involves the dephosphorylation of PLP by PDXP (Jang et al., 2003 [PubMed 14522954]).[supplied by OMIM, Mar 2008]

Uniprot Description

PDXP: Protein serine phosphatase that dephosphorylates 'Ser-3' in cofilin and probably also dephosphorylates phospho-serine residues in DSTN. Regulates cofilin-dependent actin cytoskeleton reorganization. Required for normal progress through mitosis and normal cytokinesis. Does not dephosphorylate phospho-threonines in LIMK1. Does not dephosphorylate peptides containing phospho- tyrosine. Pyridoxal phosphate phosphatase. Has some activity towards pyridoxal 5'-phosphate (PLP), pyridoxine 5'-phosphate (PMP) and pyridoxine 5'-phosphate (PNP), with a highest activity with PLP followed by PNP. Homodimer. Ubiquitous. Highly expressed in all the regions of central nerve system except the spinal cord. Also expressed at high level in liver and testis. In fetus, it is weakly expressed in all organs except brain. Inhibited by NaF, Zn(2+), Ca(2+), Mn(2+) and EDTA. Belongs to the HAD-like hydrolase superfamily.

Protein type: EC 3.1.3.74; Cofactor and Vitamin Metabolism - vitamin B6; EC 3.1.3.3; Cell cycle regulation; Protein phosphatase, Ser/Thr (non-receptor)

Chromosomal Location of Human Ortholog: 22q12.3

Cellular Component: lamellipodium; plasma membrane; midbody; cytosol; cleavage furrow; actin cytoskeleton

Molecular Function: protein binding; phosphoserine phosphatase activity; heat shock protein binding; magnesium ion binding; phosphoprotein phosphatase activity; pyridoxal phosphatase activity

Biological Process: actin rod formation; positive regulation of actin filament depolymerization; regulation of mitosis; pyridoxal phosphate catabolic process; protein amino acid dephosphorylation; regulation of cytokinesis

Product Notes

The Bovine PDXP pdxp (Catalog #AAA752557) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA752557 ELISA Kit recognizes Bovine PDXP. It is sometimes possible for the material contained within the vial of "Pyridoxal /pyridoxine vitamin B6 -phosphatase, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
