Principle of the Assay: PDXP ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PDXP antibody and an PDXP-HRP conjugate. The assay sample and buffer are incubated together with PDXP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PDXP concentration since PDXP from samples and PDXP-HRP conjugate compete for the anti-PDXP antibody binding site. Since the number of sites is limited, as more sites are occupied by PDXP from the sample, fewer sites are left to bind PDXP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PDXP concentration in each sample is interpolated from this standard curve.

Pyridoxal 5-prime-phosphate (PLP) is the active form of vitamin B6 that acts as a coenzyme in maintaining biochemical homeostasis. The preferred degradation route from PLP to 4-pyridoxic acid involves the dephosphorylation of PLP by PDXP (Jang et al., 2003 [PubMed 14522954]).[supplied by OMIM, Mar 2008]

Regulates cofilin-dependent actin cytoskeleton reorganization. Required for normal progress through mitosis and normal cytokinesis. Does not dephosphorylate phospho-threonines in LIMK1. Does not dephosphorylate peptides containing phospho- tyrosine. Pyridoxal phosphate phosphatase. Has some activity towards pyridoxal 5'-phosphate (PLP), pyridoxine 5'-phosphate (PMP) and pyridoxine 5'-phosphate (PNP), with a highest activity with PLP followed by PNP. Homodimer. Ubiquitous.
