INTENDED USE This PDPN ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine PDPN. This ELISA kit for research use only!
PRINCIPLE OF THE ASSAY PDPN ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PDPN antibody and an PDPN-HRP conjugate. The assay sample and buffer are incubated together with PDPN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PDPN concentration since PDPN from samples and PDPN-HRP conjugate compete for the anti-PDPN antibody binding site. Since the number of sites is limited, as more sites are occupied by PDPN from the sample, fewer sites are left to bind PDPN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PDPN concentration in each sample is interpolated from this standard curve.

This gene encodes a type-I integral membrane glycoprotein with diverse distribution in human tissues. The physiological function of this protein may be related to its mucin-type character. The homologous protein in other species has been described as a differentiation antigen and influenza-virus receptor. The specific function of this protein has not been determined but it has been proposed as a marker of lung injury. Required for normal lung cell proliferation and alveolus formation at birth. Induces platelet aggregation. Does not have any effect on folic acid or amino acid transport. Does not function as a water channel or as a regulator of aquaporin-type water channels. Belongs to the podoplanin family. 4 isoforms of the human protein are produced by alternative splicing.
