Principle of the Assay: OTX1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-OTX1 antibody and an OTX1-HRP conjugate. The assay sample and buffer are incubated together with OTX1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the OTX1 concentration since OTX1 from samples and OTX1-HRP conjugate compete for the anti-OTX1 antibody binding site. Since the number of sites is limited, as more sites are occupied by OTX1 from the sample, fewer sites are left to bind OTX1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The OTX1 concentration in each sample is interpolated from this standard curve.

This gene encodes a precursor protein that is processed to produce oxytocin and neurophysin I. Oxytocin is a posterior pituitary hormone which is synthesized as an inactive precursor in the hypothalamus along with its carrier protein neurophysin I. Together with neurophysin, it is packaged into neurosecretory vesicles and transported axonally to the nerve endings in the neurohypophysis, where it is either stored or secreted into the bloodstream. The precursor seems to be activated while it is being transported along the axon to the posterior pituitary. This hormone contracts smooth muscle during parturition and lactation. It is also involved in cognition, tolerance, adaptation and complex sexual and maternal behaviour, as well as in the regulation of water excretion and cardiovascular functions.
