Intended Uses: This NXF1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Chicken NXF1. This ELISA kit for research use only!

Principle of the Assay: NXF1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-NXF1 antibody and an NXF1-HRP conjugate. The assay sample and buffer are incubated together with NXF1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NXF1 concentration since NXF1 from samples and NXF1-HRP conjugate compete for the anti-NXF1 antibody binding site. Since the number of sites is limited, as more sites are occupied by NXF1 from the sample, fewer sites are left to bind NXF1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NXF1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene is one member of a family of nuclear RNA export factor genes. Common domain features of this family are a noncanonical RNP-type RNA-binding domain (RBD), 4 leucine-rich repeats (LRRs), a nuclear transport factor 2 (NTF2)-like domain that allows heterodimerization with NTF2-related export protein-1 (NXT1), and a ubiquitin-associated domain that mediates interactions with nucleoporins. The LRRs and NTF2-like domains are required for export activity. Alternative splicing seems to be a common mechanism in this gene family. The encoded protein of this gene shuttles between the nucleus and the cytoplasm and binds in vivo to poly(A)+ RNA. It is the vertebrate homologue of the yeast protein Mex67p. The encoded protein overcomes the mRNA export block caused by the presence of saturating amounts of CTE (constitutive transport element) RNA of type D retroviruses. Belongs to the NXF family. 2 isoforms of the human protein are produced by alternative splicing.
