Principle of the Assay: MBS Rat TRK A ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Rat TRK A in Cell Culture Supernatants, Cell Lysates, Tissue Homogenates. This assay employs an antibody specific for Rat TRK A coated on a 96-well plate. Standards and samples are pipetted into the wells and TRK A present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Rat TRK A antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TRK A bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: Tropomyosin receptor kinase A (TrkA) are efficacious in attenuating skeletal pain. TrkA mutants were able to activate signaling cascades and were even more efficient in promoting neurite outgrowth than the wild-type receptor. TrkA is part of a sub-family of protein kinases which includes TrkB and TrkC. Also, there are other neurotrophic factors structurally related to NGF: BDNF (for Brain-Derived Neurotrophic Factor), NT-3 (for Neurotrophin-3) and NT-4 (for Neurotrophin-4). While TrkA mediates the effects of NGF, TrkB is bound and activated by BDNF, NT-4, and NT-3. Further, TrkC binds and is activated by NT-3. TrkA receptor was found in kehumanoconus-affected corneas, along with an increased level of repressor isoform of Sp3 transcription factor.

NCBI Description

a protooncogene; receptor that binds to nerve growth factor (NGF) [RGD, Feb 2006]

Uniprot Description

TrkA: a receptor tyrosine kinase of the Trk family. High affinity receptor for nerve growth factor, neurotrophin-3 and neurotrophin-4/5 but not brain- derived neurotrophic factor. Known substrates include Shc, PI-3K, and PLC-gamma-1. Has a crucial role in the development and function of the nociceptive reception system. Two splice variant isoforms have been described.

Product Notes

The Rat NTRK1 ntrk1 (Catalog #AAA824745) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA824745 ELISA Kit recognizes Rat NTRK1. AAA Biotech's TRK A can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (SE). Researchers should empirically determine the suitability of the NTRK1 ntrk1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TRK A, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
