Host Note
Optimized DNA sequence encoding Human Neurotrophin-3 mature chain was expressed in Escherichia Coli.
Endotoxin
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/ug (1EU/ug).
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Molecular Weight Note
Human Neurotrophin-3 is generated by the proteolytic removal of the signal peptide
and propeptide, the molecule has a calculated molecular mass of approximately 14 kDa.

Recombinant NT-3 is a disulfide-linked homodimer protein consisting of two 120 amino acid residue subunits, and migrates as an approximately 27 kDa protein under non-reducing and as 14 kDa under reducing conditions in SDS-PAGE.
Preparation and Storage
The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C.
Upon reconstitution, this cytokine can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.
Related Product Information for NT-3 recombinant protein
NT-3 is found in neurons of the central nervous system. NT-3 is expressed also in muscles and its expression is down-regulated in denervated muscles. Many human gliomas express and secrete NT-3 into the conditioned medium.ome protein domains of NT-3 are identical with those of NGF and BDNF.NT-3 selectively supports the survival of neuronal cell populations. NT-3 has been shown recently to prevent death of cultured embryonic rat spinal motor neurons at picomolar concentrations. NT-3 has been shown to enhance sprouting of corticospinal tract during development and after adult spinal cord lesion. The activities of NT-3 and BDNF are additive in some systems. The biological activities of NT- 3 are mediated by a receptor belonging to the trk family of receptors with intrinsic tyrosine-specific protein kinase activity. NT-3 only binds weakly to the trk receptor which is a high- affinity receptor for NGF.

NCBI Description

The protein encoded by this gene is a member of the neurotrophin family, that controls survival and differentiation of mammalian neurons. This protein is closely related to both nerve growth factor and brain-derived neurotrophic factor. It may be involved in the maintenance of the adult nervous system, and may affect development of neurons in the embryo when it is expressed in human placenta. NTF3-deficient mice generated by gene targeting display severe movement defects of the limbs. The mature peptide of this protein is identical in all mammals examined including human, pig, rat and mouse. [provided by RefSeq, Jul 2008]

Uniprot Description

NT-3: Seems to promote the survival of visceral and proprioceptive sensory neurons. Belongs to the NGF-beta family. 2 isoforms of the human protein are produced by alternative splicing.

It is sometimes possible for the material contained within the vial of "Neurotrophin-3, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
