Preparation and Storage
Storage: Avoid repeated freeze/thaw cycles.
Store at 2-8 degree C for one month.
Aliquot and store at -80 degree C for 12 months.
Stability Test: The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37 degree C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.

NCBI Description

This gene encodes a member of the nuclear receptor superfamily, and is a key regulator of xenobiotic and endobiotic metabolism. The protein binds to DNA as a monomer or a heterodimer with the retinoid X receptor and regulates the transcription of target genes involved in drug metabolism and bilirubin clearance, such as cytochrome P450 family members. Unlike most nuclear receptors, this transcriptional regulator is constitutively active in the absence of ligand but is regulated by both agonists and inverse agonists. Ligand binding results in translocation of this protein to the nucleus, where it activates or represses target gene transcription. These ligands include bilirubin, a variety of foreign compounds, steroid hormones, and prescription drugs. Multiple transcript variants encoding different isoforms have been found for this gene. Uniprot Description

NR1I3: Binds and transactivates the retinoic acid response elements that control expression of the retinoic acid receptor beta 2 and alcohol dehydrogenase 3 genes. Transactivates both the phenobarbital responsive element module of the human CYP2B6 gene and the CYP3A4 xenobiotic response element. Interacts with ECT2. Heterodimer of NR1I3 and RXR. Interacts with PSMC4. Directly interacts with DNAJC7. The DNAJC7-NR1I3 complex may also include HSP90. By dexamethasone. Predominantly expressed in liver. Belongs to the nuclear hormone receptor family. NR1 subfamily. 7 isoforms of the human protein are produced by alternative splicing.

Protein type: Nuclear receptor; DNA-binding

Chromosomal Location of Human Ortholog: 1q23.3

Cellular Component: cytoplasm; cytoskeleton; cytosol; nucleoplasm; nucleus

Molecular Function: androgen receptor activity; DNA binding; ligand-dependent nuclear receptor activity; thyroid hormone receptor activity; transcription coactivator activity; transcription factor activity; zinc ion binding

Biological Process: androgen receptor signaling pathway; gene expression; intracellular receptor-mediated signaling pathway; negative regulation of transcription, DNA-dependent; osteoblast differentiation; positive regulation of transcription from RNA polymerase II promoter; signal transduction; steroid hormone mediated signaling; transcription initiation from RNA polymerase II promoter

Product Notes

The CAR nr1i3 (Catalog #AAA2033523) is a Recombinant Protein produced from E. coli and is intended for research purposes only. The product is available for immediate purchase. The Recombinant Constitutive Androstane Receptor (CAR) reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's Constitutive Androstane Receptor can be used in a range of immunoassay formats including, but not limited to, Positive Control, Immunogen, SDS-PAGE, WB. (May be suitable for use in other assays to be determined by the end user.) . Researchers should empirically determine the suitability of the CAR nr1i3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Constitutive Androstane Receptor, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
