Canine Nuclear factor NF-kappa-B p105 subunit ELISA Kit
Principle of the Assay: NFKB1/p105 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-NFKB1/p105 antibody and an NFKB1/p105-HRP conjugate. The assay sample and buffer are incubated together with NFKB1/p105-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NFKB1/p105 concentration since NFKB1/p105 from samples and NFKB1/p105-HRP conjugate compete for the anti-NFKB1/p105 antibody binding site. Since the number of sites is limited, as more sites are occupied by NFKB1/p105 from the sample, fewer sites are left to bind NFKB1/p105-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NFKB1/p105 concentration in each sample is interpolated from this standard curve.

This gene encodes a 105 kD protein which can undergo cotranslational processing by the 26S proteasome to produce a 50 kD protein. The 105 kD protein is a Rel protein-specific transcription inhibitor and the 50 kD protein is a DNA binding subunit of the NF-kappa-B (NFKB) protein complex. NFKB is a transcription regulator that is activated by various intra- and extra-cellular stimuli such as cytokines, oxidant-free radicals, ultraviolet irradiation, and bacterial or viral products. Activated NFKB translocates into the nucleus and stimulates the expression of genes involved in a wide variety of biological functions. Inappropriate activation of NFKB has been associated with a number of inflammatory diseases while persistent inhibition of NFKB leads to inappropriate immune cell development or delayed cell growth.
