Intended Uses: This NCL ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Chicken NCL. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: NCL ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-NCL antibody and an NCL-HRP conjugate. The assay sample and buffer are incubated together with NCL-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NCL concentration since NCL from samples and NCL-HRP conjugate compete for the anti-NCL antibody binding site. Since the number of sites is limited, as more sites are occupied by NCL from the sample, fewer sites are left to bind NCL-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NCL concentration in each sample is interpolated from this standard curve.

NCBI Description

Nucleolin (NCL), a eukaryotic nucleolar phosphoprotein, is involved in the synthesis and maturation of ribosomes. It is located mainly in dense fibrillar regions of the nucleolus. Human NCL gene consists of 14 exons with 13 introns and spans approximately 11kb. The intron 11 of the NCL gene encodes a small nucleolar RNA, termed U20. [provided by RefSeq, Jul 2008]

It induces chromatin decondensation by binding to histone H1. It is thought to play a role in pre-rRNA transcription and ribosome assembly. May play a role in the process of transcriptional elongation. Binds RNA oligonucleotides with 5'-UUAGGG-3' repeats more tightly than the telomeric single-stranded DNA 5'-TTAGGG-3' repeats.
