Form/Format
Recombinant Myostatin was lyophilized from a 0.2 um filtered Tris solution pH8.0.
Applicable Applications for MSTN recombinant protein
Cell Assay
Species
Human
Endotoxin Level
<1.0EU/ug of recombinant protein as determined by the LAL method
Sample Cell Assay Data
The ED(50) was determined by the dose-dependent proliferation inhibition of human MPC-11 cells was found to be in the range of 20-40ng/mL.
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1mg/ml. This solution can then be diluted into other buffers.
Preparation and Storage
The lyophilized protein is stable for at least one year from date of receipt at -70 degree C. Upon reconstitution, this cytokine can be stored in working aliquots at 2 degree -8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity. Avoid repeated freeze/thaw cycles.

Recombinant Human MSTN was expressed in E Coli cells. The protein accession number is O14793
Growth differentiation Factor 8 (GDF-8), also known as myostatin, is a secreted protein that is expressed specifically in developing and adult skeletal muscle. It controls myoblast proliferation and is a potent negative regulator of skeletal muscle mass. GDF-8 belongs to the transforming growth factor beta (TGF-beta) superfamily, which includes the TGF-betas, bone morphogenetic proteins (BMPs), growth differentiation factors (GDFs), activins, inhibins, leftys, nodal, Mullerian inhibitory substance (MIS) and the glial cell line-derived neurotrophic factors (GDNFs). All TGF-beta superfamily members are synthesized and secreted as a homodimeric prepropeptide that is cleaved by proprotein convertases such as furin to generate the dimeric N-terminal propeptide and the dimeric C-terminal mature active protein.

Uniprot Description

MSTN: Acts specifically as a negative regulator of skeletal muscle growth. Defects in MSTN are the cause of muscle hypertrophy (MSLHP). MSLHP is a condition characterized by increased muscle bulk and strength. Affected individuals are exceptionally strong. Belongs to the TGF-beta family.

Protein type: Secreted; Secreted, signal peptide

Chromosomal Location of Human Ortholog: 2q32.2

Cellular Component: extracellular space

Molecular Function: cytokine activity; identical protein binding; protein binding; receptor binding; transforming growth factor beta receptor binding

Biological Process: cell development; muscle development; muscle maintenance; negative regulation of myoblast differentiation; negative regulation of protein kinase B signaling cascade; negative regulation of skeletal muscle growth; positive regulation of transcription, DNA-dependent; regulation of apoptosis; regulation of MAPKKK cascade

Product Notes

The MSTN mstn (Catalog #AAA517033) is a Recombinant Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's MSTN can be used in a range of immunoassay formats including, but not limited to, Cell Assay. Researchers should empirically determine the suitability of the MSTN mstn for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MSTN, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
