Principle of the Assay: ZIC1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ZIC1 antibody and an ZIC1-HRP conjugate. The assay sample and buffer are incubated together with ZIC1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ZIC1 concentration since ZIC1 from samples and ZIC1-HRP conjugate compete for the anti-ZIC1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ZIC1 from the sample, fewer sites are left to bind ZIC1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ZIC1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the ZIC family of C2H2-type zinc finger proteins. Members of this family are important during development. Aberrant expression of this gene is seen in medulloblastoma, a childhood brain tumor. This gene is closely linked to the gene encoding zinc finger protein of the cerebellum 4, a related family member on chromosome 3. This gene encodes a transcription factor that can bind and transactivate the apolipoprotein E gene. [provided by RefSeq, Jul 2008]

Promotes MF migration towards ipsilaterally- located cerebellar territories. May have a role in shear flow mechanotransduction in osteocytes. Retains nuclear GLI1 and GLI3 in the cytoplasm. Binds to the minimal GLI-consensus sequence 5'- TGGGTGGTC-3'. Belongs to the GLI C2H2-type zinc-finger protein family.
