Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Mouse Tryptophan 5-hydroxylase 2 (TPH2) ELISA Kit employs a two-site sandwich ELISA to quantitate TPH2 in samples. An antibody specific for TPH2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TPH2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TPH2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TPH2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: TPH2 encodes a member of the pterin-dependent aromatic acid hydroxylase family. The encoded protein catalyzes the first and rate limiting step in the biosynthesis of serotonin, an important hormone and neurotransmitter. The human genome contains two related tryptophan hydroxylases, one on chromosome 11p15-p14 and one on chromosome 12q21. This gene is expressed predominantly in the brain stem. Mutations in this gene may be associated with psychiatric diseases such as bipolar affective disorder and major depression. This RefSeq record was created from transcript and genomic sequence data because no single transcript was available for the full length of the gene. The extent of this transcript is supported by the mouse ortholog.

Uniprot Description

TPH2: an enzyme that is rate-limiting in the biosynthesis of serotonin. It catalyzes the biopterin dependent monooxygenation of tryptophan to 5-hydroxytryptophan (5HT), which is subsequently decarboxylated to form the neurotransmitter serotonin. Its expression is limited to a few specialized tissues: raphe neurons, pinealocytes, mast cells, mononuclear leukocytes, beta-cells of the islets of Langerhans, and intestinal and pancreatic enterochromaffin cells. Two alternatively spliced isoforms have been described. Isoform 2 appears to be expressed exclusively in the brain, and is expressed at significantly higher levels in the raphe nuclei than isoform 1.
