Intended Uses: This MAFF ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse MAFF. This ELISA kit for research use only!

Principle of the Assay: MAFF ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-MAFF antibody and an MAFF-HRP conjugate. The assay sample and buffer are incubated together with MAFF-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MAFF concentration since MAFF from samples and MAFF-HRP conjugate compete for the anti-MAFF antibody binding site. Since the number of sites is limited, as more sites are occupied by MAFF from the sample, fewer sites are left to bind MAFF-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MAFF concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a basic leucine zipper (bZIP) transcription factor that lacks a transactivation domain. It is known to bind the US-2 DNA element in the promoter of the oxytocin receptor (OTR) gene and most likely heterodimerizes with other leucine zipper-containing proteins to enhance expression of the OTR gene during term pregnancy. The encoded protein can also form homodimers, and since it lacks a transactivation domain, the homodimer may act as a repressor of transcription. This gene may also be involved in the cellular stress response. Multiple transcript variants encoding two different isoforms have been found for this gene. [provided by RefSeq, Jun 2009]

MAFF: Interacts with the upstream promoter region of the oxytocin receptor gene. May be a transcriptional enhancer in the up-regulation of the oxytocin receptor gene at parturition. Since it lacks a putative transactivation domain, it may behave as a transcriptional repressor when it dimerize among himself. May also serve as a transcriptional activator by dimerizing with other (usually larger) basic-zipper proteins and recruiting them to specific DNA-binding sites. May be involved in the cellular stress response. Belongs to the bZIP family. Maf subfamily. 2 isoforms of the human protein are produced by alternative splicing.
