Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-TPO polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-TPO polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the TPO amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of TPO can be calculated.

Background: Thrombopoietin (THPO) also known as megakaryocyte growth and development factor (MGDF) is a protein that in humans is encoded by the THPO gene. The gene is 6.2 kb long and contains 6 exons and 5 introns, and maps to 3q27. THPO was expressed from both promoters in HEK293T human embryonic kidney cells and in HepG2 adult liver cells. The protein encoded by this gene is a humoral growth factor that is necessary for megakaryocyte proliferation and maturation, as well as for thrombopoiesis. This protein is the ligand for MLP/C_MPL, the product of myeloproliferative leukemia virus oncogene.

NCBI Description

Megakaryocytopoiesis is the cellular development process that leads to platelet production. The protein encoded by this gene is a humoral growth factor that is necessary for megakaryocyte proliferation and maturation, as well as for thrombopoiesis. This protein is the ligand for MLP/C_MPL, the product of myeloproliferative leukemia virus oncogene. Mutations in this gene are the cause of thrombocythemia 1. Alternate splicing results in multiple transcript variants of this gene.[provided by RefSeq, Jun 2012]

Uniprot Description

Function: Lineage-specific cytokine affecting the proliferation and maturation of megakaryocytes from their committed progenitor cells. It acts at a late stage of megakaryocyte development. It may be the major physiological regulator of circulating platelets.

Subcellular location: Secreted.

Domain: Two-domain structure with an erythropoietin-like N-terminal and a Ser/Pro/Thr-rich C-terminal.

Involvement in disease: Thrombocythemia 1 (THCYT1) [MIM:187950]: A myeloproliferative disorder characterized by excessive platelet production, resulting in increased numbers of circulating platelets. It can be associated with spontaneous hemorrhages and thrombotic episodes.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.10

Sequence similarities: Belongs to the EPO/TPO family.

Product Notes

The Mouse TPO thpo (Catalog #AAA355419) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA355419 ELISA Kit recognizes Mouse TPO. AAA Biotech's Thrombopoietin / TPO can be used in a range of immunoassay formats including, but not limited to, Quantitative sELISA (EIA). For quantitative detection of TPO in mouse serum, body fluids, tissue lysates or cell culture supernatants. Researchers should empirically determine the suitability of the TPO thpo for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Thrombopoietin / TPO, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
