Intended Uses: This ELISA kit applies to the in vitro quantitative determination of Mouse Thrombopoietin antibody concentrations in serum, plasma and other biological fluids.

Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antigen specific to Thrombopoietin antibody. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antigen. Then a biotinylated detection antigen specific for Thrombopoietin antibody and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Thrombopoietin antibody, biotinylated detection antigen and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of Thrombopoietin antibody.You can calculate the concentration of Thrombopoietin antibody in the samples by comparing the OD of the samples to the standard curve.

NCBI Description

Megakaryocytopoiesis is the cellular development process that leads to platelet production. The main functional protein encoded by this gene is a humoral growth factor that is necessary for megakaryocyte proliferation and maturation, as well as for thrombopoiesis. This protein is the ligand for MLP/C_MPL, the product of myeloproliferative leukemia virus oncogene. Mutations in this gene are the cause of thrombocythemia 1. Alternative promoter usage and differential splicing result in multiple transcript variants differing in the 5' UTR and/or coding region. Multiple AUG codons upstream of the main open reading frame (ORF) have been identified, and these upstream AUGs inhibit translation of the main ORF at different extent. THPO: Lineage-specific cytokine affecting the proliferation and maturation of megakaryocytes from their committed progenitor cells. It acts at a late stage of megakaryocyte development. It may be the major physiological regulator of circulating platelets. Defects in THPO are the cause of thrombocythemia type 1 (THCYT1). A myeloproliferative disorder characterized by elevated platelet levels due to sustained proliferation of megakaryocytes, and frequently lead to thrombotic and haemorrhagic complications. Belongs to the EPO/TPO family. 3 isoforms of the human protein are produced by alternative splicing.
