Intended Uses: This PSME1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse PSME1. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: PSME1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-PSME1 antibody and an PSME1-HRP conjugate. The assay sample and buffer are incubated together with PSME1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PSME1 concentration since PSME1 from samples and PSME1-HRP conjugate compete for the anti-PSME1 antibody binding site. Since the number of sites is limited, as more sites are occupied by PSME1 from the sample, fewer sites are left to bind PSME1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PSME1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The 26S proteasome is a multicatalytic proteinase complex with a highly ordered structure composed of 2 complexes, a 20S core and a 19S regulator. The 20S core is composed of 4 rings of 28 non-identical subunits; 2 rings are composed of 7 alpha subunits and 2 rings are composed of 7 beta subunits. The 19S regulator is composed of a base, which contains 6 ATPase subunits and 2 non-ATPase subunits, and a lid, which contains up to 10 non-ATPase subunits. Proteasomes are distributed throughout eukaryotic cells at a high concentration and cleave peptides in an ATP/ubiquitin-dependent process in a non-lysosomal pathway. An essential function of a modified proteasome, the immunoproteasome, is the processing of class I MHC peptides. The immunoproteasome contains an alternate regulator, referred to as the 11S regulator or PA28, that replaces the 19S regulator. Three subunits (alpha, beta and gamma) of the 11S regulator have been identified. This gene encodes the alpha subunit of the 11S regulator, one of the two 11S subunits that is induced by gamma-interferon. Three alpha and three beta subunits combine to form a heterohexameric ring.
