The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Platelet/Endothelial Cell Adhesion Molecule (PECAM1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Platelet/Endothelial Cell Adhesion Molecule (PECAM1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Platelet/Endothelial Cell Adhesion Molecule (PECAM1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Platelet/Endothelial Cell Adhesion Molecule (PECAM1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The protein encoded by this gene is found on the surface of platelets, monocytes, neutrophils, and some types of T-cells, and makes up a large portion of endothelial cell intercellular junctions. Tyr-690 plays a critical role in TEM and is required for efficient trafficking of PECAM1 to and from the lateral border recycling compartment (LBRC) and is also essential for the LBRC membrane to be targeted around migrating leukocytes (PubMed:19342684). Heterophilic interaction with CD177 plays a role in transendothelial migration of neutrophils (PubMed:17580308).
