Mouse Metabotropic glutamate receptor 2 (GRM2) ELISA Kit
Principle of the Assay: GRM2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-GRM2 antibody and an GRM2-HRP conjugate. The assay sample and buffer are incubated together with GRM2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GRM2 concentration since GRM2 from samples and GRM2-HRP conjugate compete for the anti-GRM2 antibody binding site. Since the number of sites is limited, as more sites are occupied by GRM2 from the sample, fewer sites are left to bind GRM2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GRM2 concentration in each sample is interpolated from this standard curve.

L-glutamate is the major excitatory neurotransmitter in the central nervous system and activates both ionotropic and metabotropic glutamate receptors. Glutamatergic neurotransmission is involved in most aspects of normal brain function and can be perturbed in many neuropathologic conditions. The metabotropic glutamate receptors are a family of G protein-coupled receptors, that have been divided into 3 groups on the basis of sequence homology, putative signal transduction mechanisms, and pharmacologic properties. Group I includes GRM1 and GRM5 and these receptors have been shown to activate phospholipase C. Group II includes GRM2 and GRM3 while Group III includes GRM4, GRM6, GRM7 and GRM8. Group II and III receptors are linked to the inhibition of the cyclic AMP cascade but differ in their agonist selectivities.
