Mouse Glutamate carboxypeptidase 2 (FOLH1) ELISA Kit
Principle of the Assay: FNDC1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-FNDC1 antibody and an FNDC1-HRP conjugate. The assay sample and buffer are incubated together with FNDC1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FNDC1 concentration since FNDC1 from samples and FNDC1-HRP conjugate compete for the anti-FNDC1 antibody binding site. Since the number of sites is limited, as more sites are occupied by FNDC1 from the sample, fewer sites are left to bind FNDC1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FNDC1 concentration in each sample is interpolated from this standard curve.

This gene encodes a type II transmembrane glycoprotein belonging to the M28 peptidase family. The protein acts as a glutamate carboxypeptidase on different alternative substrates, including the nutrient folate and the neuropeptide N-acetyl-l-aspartyl-l-glutamate and is expressed in a number of tissues such as prostate, central and peripheral nervous system and kidney. A mutation in this gene may be associated with impaired intestinal absorption of dietary folates, resulting in low blood folate levels and consequent hyperhomocysteinemia. Expression of this protein in the brain may be involved in a number of pathological conditions associated with glutamate excitotoxicity. In the prostate the protein is up-regulated in cancerous cells and is used as an effective diagnostic and prognostic indicator of prostate cancer. This gene likely arose from a duplication event of a nearby chromosomal region. Alternative splicing gives rise to multiple transcript variants encoding several different isoforms. Isoform PSM-4 and isoform PSM-5 would appear to be physiologically irrelevant. Involved in prostate tumor progression. Belongs to the peptidase M28 family. M28B subfamily. 8 isoforms of the human protein are produced by alternative splicing.
