Mouse Epididymal secretory glutathione peroxidase (GPX5) ELISA Kit
Principle of the Assay: GPX5 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-GPX5 antibody and an GPX5-HRP conjugate. The assay sample and buffer are incubated together with GPX5-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GPX5 concentration since GPX5 from samples and GPX5-HRP conjugate compete for the anti-GPX5 antibody binding site. Since the number of sites is limited, as more sites are occupied by GPX5 from the sample, fewer sites are left to bind GPX5-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GPX5 concentration in each sample is interpolated from this standard curve.

This gene belongs to the glutathione peroxidase family. It is specifically expressed in the epididymis in the mammalian male reproductive tract, and is androgen-regulated. Unlike mRNAs for other characterized glutathione peroxidases, this mRNA does not contain a selenocysteine (UGA) codon. Thus, the encoded protein is selenium-independent, and has been proposed to play a role in protecting the membranes of spermatozoa from the damaging effects of lipid peroxidation and/or preventing premature acrosome reaction. Alternatively spliced transcript variants encoding different isoforms have been described for this gene. [provided by RefSeq, Oct 2008]
