Mouse DNA topoisomerase 2-alpha (TOP2A) ELISA Kit
Principle of the Assay: TOP2A ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-TOP2A antibody and an TOP2A-HRP conjugate. The assay sample and buffer are incubated together with TOP2A-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TOP2A concentration since TOP2A from samples and TOP2A-HRP conjugate compete for the anti-TOP2A antibody binding site. Since the number of sites is limited, as more sites are occupied by TOP2A from the sample, fewer sites are left to bind TOP2A-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TOP2A concentration in each sample is interpolated from this standard curve.

This gene encodes a DNA topoisomerase, an enzyme that controls and alters the topologic states of DNA during transcription. This nuclear enzyme is involved in processes such as chromosome condensation, chromatid separation, and the relief of torsional stress that occurs during DNA transcription and replication. It catalyzes the transient breaking and rejoining of two strands of duplex DNA which allows the strands to pass through one another, thus altering the topology of DNA. Two forms of this enzyme exist as likely products of a gene duplication event. The gene encoding this form, alpha, is localized to chromosome 17 and the beta gene is localized to chromosome 3. The gene encoding this enzyme functions as the target for several anticancer agents and a variety of mutations in this gene have been associated with the development of drug resistance. Reduced activity of this enzyme may also play a role in ataxia-telangiectasia. [provided by RefSeq, Jul 2010]
