Reactivity
Mouse
Specificity
This assay has high sensitivity and excellent specificity for detection of DSTN. No significant cross-reactivity or interference between DSTN and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between DSTN and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Sensitivity
1.0 pg/mL

Intended Uses: This DSTN ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse DSTN. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: DSTN ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-DSTN antibody and an DSTN-HRP conjugate. The assay sample and buffer are incubated together with DSTN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DSTN concentration since DSTN from samples and DSTN-HRP conjugate compete for the anti-DSTN antibody binding site. Since the number of sites is limited, as more sites are occupied by DSTN from the sample, fewer sites are left to bind DSTN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DSTN concentration in each sample is interpolated from this standard curve.

Uniprot Description

Destrin: an actin-depolymerizing cytoskeletal protein that belongs to the ADF actin-binding protein family. Severs actin filaments (F-actin) and binds to actin monomers (G-actin). Present in many vertebrate tissues and particularly abundant in neuronal cells. It binds F-actin in a pH-dependent manner which affects it's severing/depolymerizing activities. It also binds G-actin at pH 8.0 thereby sequestering monomers and preventing polymerization.

Protein type: Actin-binding; Motility/polarity/chemotaxis

Chromosomal Location of Human Ortholog: 20p12.1

Cellular Component: cortical actin cytoskeleton; actin cytoskeleton

Molecular Function: actin binding

Biological Process: actin filament severing; actin filament depolymerization; positive regulation of actin filament depolymerization; actin polymerization and/or depolymerization

Product Notes

The Mouse DSTN dstn (Catalog #AAA7238288) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7238288 ELISA Kit recognizes Mouse DSTN. It is sometimes possible for the material contained within the vial of "Destrin (DSTN), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
