Principle of the assay: CYP4B1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CYP4B1 antibody and an CYP4B1-HRP conjugate. The assay sample and buffer are incubated together with CYP4B1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CYP4B1 concentration since CYP4B1 from samples and CYP4B1-HRP conjugate compete for the anti-CYP4B1 antibody binding site. Since the number of sites is limited, as more sites are occupied by CYP4B1 from the sample, fewer sites are left to bind CYP4B1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CYP4B1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmic reticulum. In rodents, the homologous protein has been shown to metabolize certain carcinogens; however, the specific function of the human protein has not been determined. Two transcript variants encoding slightly different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

Belongs to the cytochrome P450 family. 2 isoforms of the human protein are produced by alternative splicing.
