This assay employs a two-site sandwich ELISA to quantitate CLTA in samples. An antibody specific for CLTA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CLTA present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CLTA is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CLTA bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

Clathrin is a large, soluble protein composed of heavy and light chains. It functions as the main structural component of the lattice-type cytoplasmic face of coated pits and vesicles which entrap specific macromolecules during receptor-mediated endocytosis. This gene encodes one of two clathrin light chain proteins which are believed to function as regulatory elements. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]

Clathrin is the major protein of the polyhedral coat of coated pits and vesicles. Acts as component of the TACC3/ch-TOG/clathrin complex proposed to contribute to stabilization of kinetochore fibers of the mitotic spindle by acting as inter-microtubule bridge ().
